Tetracycline has a broad spectrum of antibiotic action. Tetracycline exhibits bacteriostatic activity against aerobic and anaerobic bacterial genera, both Gram-positive and Gram-negative. It is used in tetracycline controlled gene expression systems (gene switches) such as the tet-on and tet-off systems. It is also used to select tetracyline resistant colonies in cell culture.

Tetracycline resistance is mediated by a membrane-associated protein (tet gene), which modifies the bacterial membrane and prevents the transport of tetracycline into the cell.
